SLU-PP-332

A selective agonist of estrogen-related receptor gamma (ERRgamma), an orphan nuclear receptor involved in oxidative metabolism. Preclinical studies have investigated its potential as an exercise mimetic.

From ReVia

Mechanism of Action

SLU-PP-332 is a synthetic small molecule that selectively activates estrogen-related receptor gamma (ERRgamma), an orphan nuclear receptor highly expressed in metabolically active tissues including skeletal muscle, heart, and brown adipose tissue. ERRgamma regulates the transcription of genes involved in oxidative phosphorylation, fatty acid oxidation, and mitochondrial biogenesis. By activating ERRgamma, SLU-PP-332 has been shown in preclinical models to shift skeletal muscle fiber composition toward oxidative type I fibers, increase mitochondrial content, enhance exercise endurance, and modulate metabolic parameters — effects that partially mimic the adaptations produced by aerobic exercise training.

Research Applications

  • Studies have investigated exercise-mimetic effects on skeletal muscle fiber type composition
  • Research has examined mitochondrial biogenesis and oxidative metabolism enhancement
  • Studies have explored endurance performance effects in sedentary animal models
  • Research has assessed metabolic pathway modulation including fat oxidation and glucose handling

Key finding: Foundational work establishing the ERR family as regulators of mitochondrial metabolism and PGC-1alpha signaling, providing the biological basis for ERRgamma-targeted exercise mimetics.
